Biological Background: Alkaline Phosphatase (ALPL) Function and Localization
- Alkaline phosphatase, tissue-nonspecific isozyme (ALPL), also known as liver/bone/kidney alkaline phosphatase, is encoded by the ALPL gene and belongs to the alkaline phosphatase family.
- It functions as a hydrolase with broad substrate specificity, dephosphorylating compounds such as inorganic pyrophosphate (PPi), pyridoxal 5′-phosphate (PLP), and N-phosphocreatine. Related references: PMID:12162492, PMID:2220817
- Essential for skeletal and dental mineralization by hydrolyzing extracellular PPi, a mineralization inhibitor, to phosphate, thus promoting hydroxyapatite crystal formation. Related references: PMID:23688511, PMID:25982064
- Catalyzes the dephosphorylation of PLP to pyridoxal, the transportable form of vitamin B6, critical for neurotransmitter synthesis in the brain. Related references: PMID:20049532, PMID:2220817
- Localizes to the cell membrane via a GPI-anchor, and also associates with extracellular vesicles, mitochondrial membrane, and intermembrane space, mediating localized phosphate metabolism.
- Post-translationally modified with glycosylation, disulfide bonds, and metal binding sites for zinc and magnesium, which are essential for its catalytic activity.
- Mutations in ALPL are linked to hypophosphatasia, a metabolic bone disease, reflecting its key role in biomineralization.
- Additionally, ALPL can dephosphorylate microbial products like lipopolysaccharides (LPS), suggesting a role in modulating innate immune responses. Related references: PMID:28448526
Experimental Guidance and Technical Tips
- For Western blotting, consider a starting dilution of 1:500–1:2000 and validate in human, mouse, or rat tissue lysates that express ALPL, such as bone, liver, or kidney.
- For immunohistochemistry (IHC-P), antigen retrieval with citrate buffer may improve signal; test in known positive tissues like osteoblasts or hepatocytes.
- In immunofluorescence (IF-P), include appropriate permeabilization and validate colocalization with membrane or extracellular vesicle markers.
- For ELISA, pair with a matched detection antibody and test with recombinant ALPL protein or biological samples containing alkaline phosphatase activity.
- The immunogen spans amino acids 120–260, lying within the catalytic domain; the antibody may recognize both membrane-bound and soluble forms.
- Due to high homology among species, cross-reactivity with mouse and rat is expected, but always validate in your specific sample system.
