Biological Background: Alkaline Phosphatase (ALPL) Function and Localization

  • Alkaline phosphatase, tissue-nonspecific isozyme (ALPL), also known as liver/bone/kidney alkaline phosphatase, is encoded by the ALPL gene and belongs to the alkaline phosphatase family.
  • It functions as a hydrolase with broad substrate specificity, dephosphorylating compounds such as inorganic pyrophosphate (PPi), pyridoxal 5′-phosphate (PLP), and N-phosphocreatine. Related references: PMID:12162492, PMID:2220817
  • Essential for skeletal and dental mineralization by hydrolyzing extracellular PPi, a mineralization inhibitor, to phosphate, thus promoting hydroxyapatite crystal formation. Related references: PMID:23688511, PMID:25982064
