Biological Background: Alkaline Phosphatase (ALPL) Function and Localization
- Alkaline phosphatase, tissue-nonspecific isozyme (ALPL) is expressed in liver, bone, and kidney, as indicated by its alternative name.
- It catalyzes the hydrolysis of diphosphate (PPi) to phosphate, removing a potent inhibitor of mineralization, thus promoting hydroxyapatite crystal formation in bone and teeth. Related references: PMID:12162492, PMID:23688511, PMID:25982064
- ALPL dephosphorylates pyridoxal 5′-phosphate (PLP) to pyridoxal (PL), the transportable form of vitamin B6, essential for neurotransmitter synthesis in the brain. Related references: PMID:20049532, PMID:2220817
- The enzyme is localized at the cell membrane, extracellular vesicle membranes, and mitochondrial membranes, where it participates in diverse physiological processes.
- In thermogenic fat cells, mitochondrial ALPL mediates hydrolysis of N-phosphocreatine, driving a futile creatine cycle that dissipates energy as heat for adaptive thermogenesis (By similarity).
- It can dephosphorylate microbial products like lipopolysaccharides (LPS) and poly I:C, suggesting a role in modulating innate immune responses. Related references: PMID:28448526
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa1-100); this region may be buried or cleaved in mature enzyme. Confirm recognition of native protein in your experimental system.
- For Western blot (WB), a band at ~57 kDa (precursor) or ~80 kDa (mature, glycosylated) may be observed depending on sample preparation; consider using fresh lysates and protease inhibitors.
- In immunohistochemistry (IHC-P), antigen retrieval may be required for formalin-fixed paraffin-embedded sections; validate in ALPL-expressing tissues like bone, liver, or kidney.
- For ELISA applications, pairing with a detection antibody targeting a distinct epitope is recommended for sandwich format.
- Cross-reactivity with mouse and rat is confirmed; however, sequence homology should be verified if using in other species.
- Avoid multiple freeze-thaw cycles; aliquot and store at -20°C or -80°C as appropriate.
