Biological Background: ALK Function and Localization

  • ALK (Anaplastic lymphoma kinase, also known as CD246 or ALK1) is a neuronal receptor tyrosine kinase that is essentially and transiently expressed in specific regions of the central and peripheral nervous systems, where it plays a crucial role in the genesis and differentiation of the nervous system.

  • ALK acts as a key thinness protein: in hypothalamic neurons, it controls energy expenditure by negatively regulating white adipose tissue lipolysis and sympathetic tone to fine-tune energy homeostasis (By similarity).
  • Upon ligand binding (especially ALKAL2), ALK dimerizes and undergoes autophosphorylation on its intracellular kinase domain, triggering downstream activation of the mitogen-activated protein kinase (MAPK) pathway and other signaling cascades.

  • For immunofluorescence (IF/ICC), optimize fixation and permeabilization conditions to detect membrane-localized ALK; consider using cell lines with confirmed ALK expression.
  • In ELISA applications, the polyclonal nature may provide broad epitope coverage, but assay conditions and coating methods should be experimentally determined.
  • When using mouse samples, confirm sequence homology of the immunogen region, as cross-reactivity is declared but antibody performance may vary.
