Biological Background: ALIX Function and Localization

  • ALIX (programmed cell death 6-interacting protein), also known as AIP1, HP95, DRIP4, and MCPH29, is encoded by the PDCD6IP gene and exists in multiple isoforms due to alternative splicing.
  • It is a class E VPS protein essential for multivesicular body (MVB) biogenesis, concentrating and sorting cargo for intralumenal vesicle formation via the ESCRT pathway. Related references: PMID:14739459
  • ALIX binds to the phospholipid lysobisphosphatidic acid (LBPA) on MVB internal membranes and serves as an adapter for a subset of ESCRT-III proteins, such as CHMP4B.
  • The protein is required for cytokinesis, functioning in the abscission step through topologically equivalent membrane fission events. Related references: PMID:17556548 PMID:17853893 PMID:18641129
  • In concert with SDC1/4 and SDCBP, ALIX regulates exosome biogenesis, impacting intercellular communication. Related references: PMID:22660413
  • Subcellularly, ALIX localizes to the cytosol, melanosomes, centrosomes, tight junctions, midbody ring, and is secreted in extracellular exosomes.
  • It participates in host-virus interactions, facilitating HIV-1 budding by interacting with CHMP4B, and may play roles in apoptosis and epithelial barrier maintenance.
  • Post-translational modifications include acetylation, phosphorylation, and methylation, influencing its multifunctional activities.

Experimental Guidance and Technical Tips

  • The immunogen used for this antibody is recombinant protein; consider that the antibody may recognize both native and denatured ALIX depending on the application.
  • For Western blot (WB), validate in your specific sample system and optimize protein loading to detect the 96 kDa band; note that post-translational modifications might cause slight mobility shifts.
  • In IF/ICC experiments, fixation and permeabilization methods may affect staining of different ALIX pools (cytosolic, centrosomal, midbody); consider testing multiple protocols.
  • For IP, confirm antibody compatibility with your lysis buffer and ensure target enrichment by running parallel controls.
  • When using ELISA, include appropriate positive (recombinant protein) and negative controls to assess sensitivity and specificity.
