Biological Background: ALG5 Function and Localization
- ALG5 encodes dolichyl-phosphate beta-glucosyltransferase, also known as asparagine-linked glycosylation protein 5 homolog.
- It catalyzes the synthesis of dolichyl β-D-glucosyl phosphate (Dol-P-Glc), the glucose donor required for the terminal glucosylation steps in dolichol-linked oligosaccharide assembly.
- This enzyme functions in the N-glycosylation biosynthetic pathway, adding the glucose moiety that is subsequently transferred by ALG6, ALG8, and ALG10 to the growing dolichol-linked precursor oligosaccharide.
- ALG5 is an endoplasmic reticulum membrane protein with a signal-anchor topology, most likely active on the cytoplasmic side of the ER.
- Ubiquitously expressed, with highest levels in pancreas, placenta, liver, heart, brain, kidney, skeletal muscle, and lung.
