Biological Background: ALG2 Function and Localization

ALG2, also known as alpha-1,3/1,6-mannosyltransferase ALG2, is an endoplasmic reticulum membrane protein that catalyzes the addition of the second and third mannose residues to the dolichol-linked oligosaccharide precursor for N-glycosylation.

It sequentially adds alpha-1,3-linked mannose followed by alpha-1,6-linked mannose to produce Man3GlcNAc(2)-PP-dolichol, the substrate for ALG11. While both linkages are possible, alpha-1,3 then alpha-1,6 is the preferred route.

The dolichol-linked oligosaccharide assembly begins on the cytosolic face of the endoplasmic reticulum, where ALG2 functions, and later the glycan is flipped into the ER lumen for further processing.
