Biological Background: ALG2 Function and Localization

  • ALG2, also known as alpha-1,3/1,6-mannosyltransferase ALG2, is an endoplasmic reticulum membrane protein that catalyzes the addition of the second and third mannose residues to the dolichol-linked oligosaccharide precursor for N-glycosylation.
    Related references: PMID:12684507, PMID:35136180

  • It sequentially adds alpha-1,3-linked mannose followed by alpha-1,6-linked mannose to produce Man3GlcNAc(2)-PP-dolichol, the substrate for ALG11. While both linkages are possible, alpha-1,3 then alpha-1,6 is the preferred route.
    Related references: PMID:35136180

  • The dolichol-linked oligosaccharide assembly begins on the cytosolic face of the endoplasmic reticulum, where ALG2 functions, and later the glycan is flipped into the ER lumen for further processing.

  • ALG2 belongs to the glycosyltransferase family, and mutations in this gene have been associated with congenital disorders of glycosylation (type Ii) and congenital myasthenic syndrome.

  • The protein is annotated with alternative splicing, suggesting the existence of multiple isoforms, and proteomics identification confirms its expression in human tissues.

  • Keywords associated with ALG2 include transferase, transmembrane helix, and endoplasmic reticulum, reflecting its enzymatic role and membrane localization.

Experimental Guidance and Technical Tips

  • The immunogen used spans a central region (aa 197–416) of human ALG2, which may provide specificity for detecting the target by WB and ELISA; cross-reactivity with mouse is expected based on supplier validation.
  • For WB analysis, a band around 47 kDa is predicted; however, post-translational modifications or isoform variation could alter migration, so validate band size in your specific sample system.
  • ELISA protocols may require optimization of coating antigen and antibody dilutions; consider using recombinant ALG2 protein fragments as positive controls.
  • When working with mouse samples, verify antibody specificity, as sequence homology between human and mouse ALG2 is high in the immunogen region.
