Biological Background: ALG13 Function and Localization

  • ALG13 is the catalytic subunit of the UDP-N-acetylglucosamine transferase complex, also known as asparagine-linked glycosylation 13 homolog or glycosyltransferase 28 domain-containing protein 1. It mediates the second step of dolichol‑linked oligosaccharide biosynthesis on the cytoplasmic face of the endoplasmic reticulum.
  • Together with its obligate partner ALG14, ALG13 forms a dimeric complex that transfers a β1,4‑linked N‑acetylglucosamine (GlcNAc) from UDP‑GlcNAc to GlcNAc‑pyrophosphatedolichol, generating N,N′‑diacetylchitobiosyl diphosphodolichol. This intermediate is subsequently elongated by ALG1 and other glycosyltransferases.
  • The ALG13/ALG14 complex is anchored at the endoplasmic reticulum membrane, where the dolichol‑linked oligosaccharide precursor is assembled before flipping into the ER lumen for further processing and eventual transfer to nascent polypeptides.
  • Although annotated as a potential multifunctional enzyme based on sequence motifs (thiol protease, hydrolase domains), no glycosyltransferase or deubiquitinase activity has been detected for ALG13 outside its role in GlcNAc transfer. Its additional biochemical activities, if any, remain poorly characterized.
  • Mutations in the ALG13 gene are associated with congenital disorder of glycosylation type Is (CDG1S) and early‑infantile epileptic encephalopathies (DEE36, EIEE36), reflecting the critical importance of this enzyme in normal neurodevelopment and global protein N‑glycosylation.
  • Alternative splicing generates multiple ALG13 isoforms, and the protein is subject to ubiquitination (Ubl conjugation pathway), which may regulate its stability or function.
