  • ALDH4A1, also known as delta-1-pyrroline-5-carboxylate dehydrogenase (P5CDh) or aldehyde dehydrogenase family 4 member A1, is a mitochondrial enzyme.
  • It catalyzes the irreversible conversion of delta-1-pyrroline-5-carboxylate (P5C) to glutamate, linking proline and ornithine degradation to the urea cycle and tricarboxylic acid cycle.
  • The enzyme exhibits substrate preference for glutamic gamma-semialdehyde but can also oxidize succinic, glutaric, and adipic semialdehydes.
  • ALDH4A1 is localized to the mitochondrial matrix, where it functions in proline metabolism.
  • It is highly expressed in the liver, with moderate levels in skeletal muscle, kidney, heart, brain, placenta, lung, and pancreas.
