Biological Background: ALDH3A1 Function and Localization
- Aldehyde dehydrogenase family 3 member A1 (ALDH3A1) is a cytoplasmic enzyme belonging to the aldehyde dehydrogenase (ALDH) superfamily. It is also known as HTC-ALDH or tumor-associated aldehyde dehydrogenase.
- ALDH3A1 preferentially oxidizes aromatic aldehyde substrates and also acts on medium and long chain aldehydes, converting them into non-toxic fatty acids. Related references: PMID:2831537
- ALDHs, including ALDH3A1, are involved in the detoxification of alcohol-derived acetaldehyde and play roles in the metabolism of corticosteroids, biogenic amines, neurotransmitters, and lipid peroxidation products.
- In the cornea, ALDH3A1 constitutes about 50% of the soluble protein in epithelial cells and is thought to protect against UV-induced damage.
- The protein is a dimeric, NADP-preferring oxidoreductase. Post-translational modifications include acetylation.
- The gene Aldh3a1 (also known as Aldd, Aldh3) encodes this protein in rat.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1-103 at the N-terminus of rat ALDH3A1. This region is relatively conserved among human, mouse, and rat, which may contribute to the observed cross-reactivity.
- For Western blotting, the predicted molecular weight is approximately 50 kDa. Consider using a gel percentage appropriate for this size range and validate with a suitable loading control in your target tissue or cell line.
- For IHC and IF applications, corneal epithelium expresses high levels of ALDH3A1 and can serve as a positive control tissue. Optimize antigen retrieval and antibody dilution conditions as needed.
- In ELISA, ensure that the coating antigen or capture format is compatible with the antibody's binding characteristics. Pairing with a detection antibody should be evaluated for sensitivity and specificity.
