Biological Background: ALDH3A1 Function and Localization
- ALDH3A1 (Aldehyde dehydrogenase, dimeric NADP-preferring) is a member of the aldehyde dehydrogenase family, also known as ALDHIII or aldehyde dehydrogenase 3. It is encoded by the ALDH3A1 gene in humans.
- It plays a major role in detoxifying alcohol-derived acetaldehyde and is involved in the metabolism of corticosteroids, biogenic amines, neurotransmitters, and lipid peroxidation (Probable).
- ALDH3A1 oxidizes medium and long chain aldehydes into non-toxic fatty acids and preferentially oxidizes aromatic aldehyde substrates. Related references: PMID:1737758
- It constitutes about 50% of corneal epithelial soluble proteins and may help protect the cornea from UV-induced damage (By similarity).
- The protein localizes to the cytoplasm and is highly expressed in stomach, esophagus, and lung, with lower levels in liver and kidney.
- Keywords associated with ALDH3A1 include acetylation, lipid metabolism, NAD/NADP dependence, oxidoreductase activity, and 3D-structure availability.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant protein covering amino acids 1-300 of human ALDH3A1 (NP_001128640.1). Consider using this sequence information to verify epitope recognition or for competition assays.
- For Western blot (WB), the expected band is approximately 50kDa; include positive and negative controls from relevant tissues (e.g., cornea, stomach) to confirm specificity.
- In immunofluorescence (IF/ICC or IF-P), ALDH3A1 exhibits cytoplasmic staining; ensure proper permeabilization and use appropriate fixation methods.
- ELISA performance may be optimized by testing different coating antigen concentrations and comparing with known standard curves.
