Biological Background: ALDH1B1 Function and Localization
- ALDH1B1 (aldehyde dehydrogenase family 1 member B1, mitochondrial), also known as ALDH5, ALDHX, or nitroglycerin reductase, may contribute to the detoxification of alcohol-derived acetaldehyde and other aldehydes. Related references: PMID:20616185 PMID:25413692 PMID:35788181
- The enzyme demonstrates broad substrate specificity, preferentially oxidizing aliphatic medium-chain saturated aldehydes; it also metabolizes lipid peroxidation products 4-HNE and MDA, and exhibits in vitro nitroglycerin reductase activity. Related references: PMID:20616185 PMID:25413692
- ALDH1B1 shows an absolute preference for NAD(+) as a cofactor, with no detectable activity using NADP(+). Related references: PMID:20616185 PMID:25413692
- Subcellular localization is mitochondrial, within the matrix.
- Expression is highest in liver and testis, with lower levels in brain.
- The protein undergoes acetylation, contains a mitochondrial transit peptide, and its 3D structure has been determined; it is cataloged in the human reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 278-517) of human ALDH1B1; therefore, the antibody is expected to recognize full-length or C-terminal fragments.
- For Western blot, consider analyzing mitochondrial-enriched fractions from human liver or testis samples, where ALDH1B1 is abundantly expressed, to confirm the ~57 kDa band.
- In immunofluorescence and immunocytochemistry, mitochondrial staining is anticipated; co-staining with a mitochondrial marker is recommended to verify subcellular localization.
- As a polyclonal antibody, performance may vary between lots; validate each new lot in the relevant assay system before use.
- ELISA optimization may require testing different coating antigen concentrations and antibody dilutions.
