Biological Background: ALDH1A1 Function and Localization
- Aldehyde dehydrogenase 1A1 (ALDH1A1), also known as retinal dehydrogenase 1 (RALDH1), ALDH1, ALDC, PUMB1, or HEL-S-53e, is a cytosolic enzyme belonging to the aldehyde dehydrogenase family.
- Functions as a key enzyme in retinoic acid biosynthesis by catalyzing the irreversible oxidation of retinaldehyde to retinoic acid, downstream of retinol dehydrogenases (PubMed:25413692). This pathway is crucial for controlling teratogenic and cytotoxic levels of retinol and retinoic acid (PubMed:25413692). Related references: PMID:25413692
- Also oxidizes aldehydes from lipid peroxidation, such as (E)-4-hydroxynon-2-enal (HNE), malonaldehyde, and hexanal, thereby preventing protein adduct formation and lens opacification (PubMed:12941160, PubMed:15623782, PubMed:19296407). Related references: PMID:12941160, PMID:15623782, PMID:19296407
- Participates in the fructosamine degradation pathway by oxidizing 3-deoxyglucosone, a potent glycating agent (PubMed:17175089). Related references: PMID:17175089
- Exhibits aminobutyraldehyde dehydrogenase activity, potentially contributing to GABA biosynthesis in the midbrain (By similarity).
- Localizes primarily to the cytoplasm, cytosol, and cell projections such as axons.
- Expressed at the protein level in erythrocytes.
- Post-translationally modified by acetylation and phosphorylation; annotated with keywords including NAD binding, nucleotide-binding, and oxidoreductase activity.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide within amino acids 250-350 of human ALDH1A1; consider epitope accessibility in native conformation and perform appropriate antigen retrieval for IHC-P.
- Predicted molecular weight is 55kDa; validate the band in WB using relevant positive control lysates (e.g., human liver or retinal tissue).
- The antibody cross-reacts with mouse and rat; verify reactivity in your target species by sequence alignment of the immunogen region if needed.
- For IF/ICC, optimize fixation and permeabilization conditions to preserve the cytoplasmic localization signal.
- Suitable for ELISA; consider testing in a sandwich format with an appropriate paired antibody.
