• ALAS1 encodes 5-aminolevulinate synthase 1, a mitochondrial enzyme also known as delta-ALA synthase 1 or ALAS-H. Alternative splicing produces multiple transcript variants.
• It catalyzes the rate-limiting step in heme biosynthesis: the pyridoxal 5'-phosphate (PLP)-dependent condensation of succinyl-CoA and glycine to form 5-aminolevulinic acid (ALA).
• ALAS1 is localized to the mitochondrial inner membrane, where it functions as an acyltransferase and transferase.
• The enzyme is essential for heme production, which is required for hemoglobin, cytochromes, and other hemoproteins.
