Biological Background: AKT2 Function and Localization

  • AKT2, also designated Protein kinase B beta (PKB beta) or RAC-PK-beta, is a serine/threonine kinase belonging to the AKT kinase family (which includes AKT1, AKT2, and AKT3). These kinases are key downstream mediators of PI3K signaling, activated by various growth factors including insulin, IGF1, EGF, and PDGF.
  • A central role of AKT2 is in insulin-regulated glucose metabolism. It promotes glucose uptake by stimulating SLC2A4/GLUT4 translocation to the plasma membrane, enhances glycogen synthesis via inhibitory phosphorylation of GSK3A/B, and suppresses hepatic gluconeogenesis through CLK2 phosphorylation.
  • AKT2 supports cell survival and proliferation by phosphorylating and inactivating pro-apoptotic factors such as MAP3K5/ASK1, and by activating mTORC1 via TSC2 phosphorylation, thus regulating protein synthesis and cell growth.
