Biological Background: AKT2 Function and Localization

  • AKT2 (RAC-beta serine/threonine-protein kinase) is a member of the AKT kinase family, closely related to AKT1 and AKT3.
  • AKT kinases broadly regulate metabolism, proliferation, cell survival, growth and angiogenesis via phosphorylation of numerous downstream substrates.
  • AKT2 specifically mediates insulin-stimulated glucose uptake by promoting SLC2A4/GLUT4 translocation to the plasma membrane, and regulates glycogen synthesis by phosphorylating and inhibiting GSK3 isoforms.
  • AKT2 promotes cell survival by phosphorylating MAP3K5/ASK1 on Ser-83 (reducing oxidative stress-induced apoptosis) and by activating NF-kappa-B and CREB1 to induce transcription of pro-survival genes like BCL2 and MCL1.
  • AKT2 localizes to the cytoplasm, nucleus, cell membrane, and early endosome.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a C-terminal sequence (aa 390-490) of human AKT2. Because cross-reactivity with mouse and rat is observed, this region may be conserved; consider validating specificity in your model system.
  • In WB, the predicted band is approximately 56 kDa. Include appropriate positive/negative controls and confirm specificity via siRNA knockdown or peptide competition if needed.
  • For IHC-P and IF/ICC, optimize antigen retrieval and permeabilization steps. Note that AKT2 can be cytoplasmic, nuclear, or membrane-associated depending on cell type and stimulation.
  • For ELISA, the recombinant mAb format (rabbit monoclonal) provides high specificity and lot-to-lot consistency.
