Biological Background: AKT1 Function and Localization
- AKT1 is a serine/threonine-protein kinase, also known as Protein kinase B (PKB), belonging to the AKT kinase family (AKT1, AKT2, AKT3). Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within the C-terminal region (aa 350–480) of human AKT1, which may confer isoform specificity; however, validate with recombinant proteins or siRNA if cross-reactivity with AKT2 or AKT3 is a concern.
- For Western blotting, denaturing and reducing conditions typically reveal a band at ~56 kDa. Use appropriate positive controls (e.g., HeLa, MCF-7 lysates) and consider that observed mobility may reflect phosphorylation status.
- IF/ICC and IHC-P protocols may require permeabilization and antigen retrieval respectively. Optimize fixation methods (e.g., 4% PFA vs. methanol) for your sample type, and include blocking steps to minimize background.
- ELISA applications can be set up using indirect or sandwich formats; pairing with a suitable capture antibody may enhance sensitivity. Titrate the antibody and use a consistent coating buffer.
