Biological Background: AKR1B1 Function and Localization

  • AKR1B1 (Aldo-keto reductase family 1 member B1), also known as aldose reductase (AR), is an NADPH-dependent oxidoreductase that reduces a broad spectrum of carbonyl substrates including aldehydes, ketones, monosaccharides, bile acids, and xenobiotics. Related references: PMID:10510318, PMID:12732097, PMID:1332968
  • It catalyzes the conversion of glucose to sorbitol in the polyol pathway, which is an alternative glucose metabolic route that becomes particularly active under hyperglycemic conditions. Related references: PMID:4402695, PMID:1936586
  • AKR1B1 displays low enzymatic activity toward retinaldehyde isomers, including all-trans-retinal, 9-cis-retinal, and 13-cis-retinal. Related references: PMID:12732097, PMID:19010934, PMID:8343525
  • It reduces phospholipid aldehydes such as 1-palmitoyl-2-(5-oxovaleroyl)-sn-glycero-3-phosphoethanolamine (POVPC), which are formed during oxidative modification of phospholipids. Related references: PMID:17381426
  • The protein contributes to the detoxification of dietary and lipid-derived unsaturated carbonyls, including 4-hydroxynonenal and crotonaldehyde, as well as their glutathione-conjugates. Related references: PMID:21329684
  • AKR1B1 is primarily localized in the cytoplasm and is highly expressed in embryonic epithelial cells upon osmotic stress. Post-translational modifications include acetylation and phosphorylation, as indicated by proteomics.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 50–150 of human AKR1B1; thus, the antibody likely binds linear epitopes within this region. For western blotting, expect a main band at approximately 36 kDa and include a positive control lysate and molecular weight markers.
  • This antibody is suitable for WB, IF/ICC, and ELISA. For immunofluorescence, optimize fixation and permeabilization conditions for the target cell type and include appropriate negative controls.
  • Cross-reactivity is reported for mouse and rat, but validation in the specific experimental model is recommended.
