Biological Background: AKR1A1 Function and Localization
- Aldo-keto reductase family 1 member A1 (AKR1A1), also known as alcohol dehydrogenase [NADP+], aldehyde reductase, glucuronate reductase, and S-nitroso-CoA reductase (ScorR), is a member of the AKR superfamily that catalyzes NADPH-dependent carbonyl reduction.
- Catalyzes the NADPH-dependent reduction of a wide variety of carbonyl-containing compounds to their corresponding alcohols, with a preference for negatively charged substrates like glucuronate and succinic semialdehyde (PubMed:10510318, 30538128).

PMID:30538128 - Functions as a detoxifying enzyme, reducing toxic aldehydes such as methylglyoxal, 3-deoxyglucosone, and acrolein, which accumulate under hyperglycemic and oxidative stress conditions (By similarity).
- Acts as an inhibitor of protein S-nitrosylation by degrading S-nitroso-coenzyme A (S-nitroso-CoA) and also reduces S-nitrosoglutathione, thereby modulating metabolic reprogramming in kidney proximal tubules (PubMed:30538128, 31649033).

PMID:31649033 - Subcellular localization includes the cytoplasm, cytosol, and apical cell membrane, consistent with its role in metabolic and detoxification processes.
- Widely expressed, with high levels in kidney, salivary gland, and liver; also detected in trachea, stomach, brain, lung, prostate, placenta, mammary gland, and small intestine.
- Plays a role in the activation of procarcinogens and metabolism of xenobiotics such as anthracycline drugs (PubMed:11306097, 18276838).

PMID:18276838 - Post-translational modifications include acetylation and phosphorylation, which may regulate enzyme activity and subcellular trafficking.
Experimental Guidance and Technical Tips
- The immunogen (recombinant protein spanning amino acids 1-325) covers almost the entire protein sequence, suggesting the antibody may recognize multiple epitopes; denaturing conditions (e.g., SDS-PAGE for WB) may be required to expose linear epitopes.
- For Western blotting, a band at ~37 kDa is expected; validate specificity using AKR1A1 knockout or knockdown controls if available.
- Cross-reactivity with mouse and rat makes this antibody suitable for comparative studies, but always confirm reactivity in the relevant model organism through appropriate controls.
- AKR1A1 undergoes acetylation and phosphorylation; consider potential epitope masking and include phosphatase inhibitors or deacetylase inhibitors in lysis buffers if studying modified forms.
- For ELISA, titrate the antibody against recombinant AKR1A1 or cell/tissue lysates to determine optimal coating and detection conditions.
