Biological Background: AIRE Function and Localization

  • AIRE (Autoimmune regulator), also known as APECED protein, is a transcription factor essential for the establishment of immune self-tolerance. Mutations in the AIRE gene cause autoimmune polyendocrinopathy candidiasis ectodermal dystrophy (APECED).
  • AIRE promotes self-tolerance in the thymus by regulating the expression of tissue-restricted antigens (TRAs) in medullary thymic epithelial cells (mTECs). Related references: PMID:26084028
  • It binds to G-doublets in A/T-rich DNA, interacts with nucleosomes, and selectively recognizes histone H3 that lacks methylation at Lys-4, phosphorylation at Thr-3, and methylation at Arg-2, thereby acting as an epigenetic sensor.
  • AIRE induces the expression of thousands of TRAs, which are presented on MHC-I and MHC-II molecules to developing T-cells, enforcing central tolerance. Related references: PMID:26084028
  • In secondary lymphoid organs, a distinct population of bone marrow-derived extrathymic AIRE-expressing cells (eTACs) contributes to peripheral tolerance by inducing functional inactivation of CD4+ T-cells. Related references: PMID:23993652
  • Subcellular localization is primarily nuclear, with additional presence in the cytoplasm.
  • AIRE contains zinc-finger domains, undergoes alternative splicing (isoforms 2 and 3 are less frequently expressed), and is regulated by phosphorylation. Numerous disease-associated variants have been identified.

Experimental Guidance and Technical Tips

  • The immunogen targets the N-terminal region (amino acids 30–130); verify epitope accessibility and consider potential blocking effects if your assay involves this domain.
  • For Western blotting, endogenous AIRE runs at ~58 kDa; use thymus or mTEC-derived positive controls and include tissue-specific expression validation.
  • In IF/ICC applications, optimize fixation and permeabilization protocols to detect both nuclear and cytoplasmic pools. Validate staining in thymic tissue or relevant cell lines.
  • For ELISA, this polyclonal antibody can be used for capture or detection; pairing with a monoclonal antibody may improve specificity in sandwich formats.
