Biological Background: AIRE Function and Localization
- AIRE (Autoimmune regulator), also known as APECED protein, is a transcription factor essential for the establishment of immune self-tolerance. Mutations in the AIRE gene cause autoimmune polyendocrinopathy candidiasis ectodermal dystrophy (APECED).
- AIRE promotes self-tolerance in the thymus by regulating the expression of tissue-restricted antigens (TRAs) in medullary thymic epithelial cells (mTECs). Related references: PMID:26084028
- It binds to G-doublets in A/T-rich DNA, interacts with nucleosomes, and selectively recognizes histone H3 that lacks methylation at Lys-4, phosphorylation at Thr-3, and methylation at Arg-2, thereby acting as an epigenetic sensor.
