Biological Background: Aim2 Function and Localization
- Interferon-inducible protein AIM2 (also known as Ifi-210 or p210) is encoded by the Aim2 gene (Gm1313, Ifi210) in mouse, acting as a cytosolic sensor for double-stranded DNA.
- Serves as the primary sensor component of the AIM2 inflammasome, recognizing cytosolic double-stranded DNA (dsDNA) and triggering pyroptosis via caspase-1 (CASP1) activation and gasdermin-D (GSDMD) cleavage. Related references: PMID:19131592 PMID:19158675 PMID:19158676
- Detects dsDNA from viral and bacterial origins in a non-sequence-specific manner, mediating inflammasome assembly and interleukin-1β (IL1B) and IL18 secretion. Related references: PMID:19131592 PMID:19158675 PMID:20351692
- Mediates the DNA damage response by sensing double-strand breaks in the nucleus, leading to inflammasome assembly and pyroptosis in intestinal epithelial cells and bone marrow cells after acute ionizing radiation. Related references: PMID:27846608
- Plays a role in neurodevelopment by promoting neural cell death in response to DNA damage, thereby purging genetically compromised cells from the central nervous system. Related references: PMID:32350463
- Functions as a tumor suppressor independently of its inflammatory role, inhibiting AKT1 phosphorylation and suppressing cell proliferation in enterocytes, which reduces colorectal cancer risk. Related references: PMID:26095253 PMID:26107252 PMID:33505023
- Regulates regulatory T-cell (Treg) homeostasis by preventing AKT1 activation, thereby promoting Treg stability and restraining autoimmune responses. Related references: PMID:33505023
- Localizes to the cytoplasm, nucleus, and inflammasome complex; highly expressed in developing neurons and regulatory T-cells. Related references: PMID:27561456 PMID:33505023
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from within amino acids 50-150 of mouse Aim2, providing targeted reactivity for this polyclonal antibody.
- Recommended for Western blot (WB) and ELISA applications; consider optimizing antibody dilutions for your specific experimental setup.
- Since Aim2 expression is inducible by interferons, validate the antibody in appropriate models (e.g., interferon-treated cells or inflammation-related tissue samples).
- Cross-reactivity with rat has been reported; confirm performance in rat samples using relevant positive and negative controls.
