Biological Background: AIFM2/FSP1 Function and Localization
- AIFM2 (also known as ferroptosis suppressor protein 1, FSP1, AMID, or PRG3) is an NAD(P)H-dependent oxidoreductase acting as a key inhibitor of ferroptosis.
Related references: PMID:31634899, PMID:31634900, PMID:35922516 - At the plasma membrane, it catalyzes the reduction of coenzyme Q10 (ubiquinone-10) to ubiquinol-10, a lipophilic radical-trapping antioxidant that prevents lipid oxidative damage and ferroptosis.
Related references: PMID:31634899, PMID:31634900, PMID:37739993 - This anti-ferroptotic function operates in parallel to GPX4 and is independent of cellular glutathione levels.
Related references: PMID:31634899, PMID:31634900 - AIFM2 also exhibits warfarin-resistant vitamin K reductase activity, producing hydroquinones that serve as potent radical-trapping antioxidants.
Related references: PMID:35922516, PMID:36788244 - Under oxidative stress, AIFM2 forms a lipid adduct with 4-hydroxy-2-nonenal (HNE), translocates to the nucleus, and triggers DNA damage and cell death.
Related references: PMID:26689472 - The protein is capable of non-sequence-specific DNA binding.
Related references: PMID:15958387 - Subcellular localization includes lipid droplets, cell membrane, cytoplasm, mitochondrial membrane, and nucleus.
- Highly expressed in heart, moderately in liver and skeletal muscle, and at low levels in placenta, lung, kidney, and pancreas; expression is induced by p53/TP53.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 43-371 of human AIFM2. In Western blotting (WB), the predicted molecular weight is 41 kDa; however, endogenous expression levels may vary by tissue and cell line, so optimize loading and detection conditions.
- For immunohistochemistry (IHC-P), antigen retrieval methods and antibody dilution should be titrated; tissues with high expression (e.g., heart, liver) are suitable positive controls.
- This antibody shows cross-reactivity with mouse and rat samples, making it suitable for comparative studies; validate in your specific model system.
- When performing ELISA, consider using a recombinant protein or positive tissue lysate to establish a standard curve.
