Biological Background: AIF1/IBA1 Function and Localization

• AIF1 (Allograft inflammatory factor 1), also known as IBA1 (Ionized calcium-binding adapter molecule 1) and Protein G1, is an actin-binding protein.
• It enhances membrane ruffling and RAC activation, and promotes phagocytosis through binding actin and calcium.
• Plays a role in macrophage activation, T-lymphocyte proliferation, and lymphocyte migration, contributing to vascular inflammation.
• Localizes to the cytoplasm, cytoskeleton, and cell projections including the ruffle membrane and phagocytic cup.
• Subject to acetylation and phosphorylation, as identified by proteomics.
• Detected in T-lymphocytes and peripheral blood mononuclear cells.
