Biological Background: AIF1/IBA1 Function and Localization

  • AIF1, also known as IBA1, is an actin-binding protein with calcium-binding properties, designated as allograft inflammatory factor 1 and ionized calcium-binding adapter molecule 1.
  • It enhances membrane ruffling and RAC activation, promotes actin-bundling activity of LCP1, and plays a role in phagocytosis.
  • The protein is implicated in macrophage activation and function, promotes proliferation of vascular smooth muscle cells and T-lymphocytes, enhances lymphocyte migration, and participates in vascular inflammation.
  • Subcellularly, it localizes to the cytoplasm, cytoskeleton, cell projections, ruffling membranes, and phagocytic cups.
  • Post-translational modifications include acetylation and phosphorylation; it contains repeat regions and is part of the reference proteome.
  • Tissue expression is detected in T-lymphocytes and peripheral blood mononuclear cells.
  • The AIF1 gene (GeneID: 199) undergoes alternative splicing, generating multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1–147 of human AIF1/IBA1, covering the full-length protein and supporting recognition of native conformations.
  • For western blotting, an expected band around 17 kDa should be observed; consider validating in lysates from macrophage or lymphocyte-rich samples.
  • In immunohistochemistry (paraffin), antigen retrieval conditions and detection in tissues with known AIF1 expression, such as inflammatory lesions, should be optimized.
  • For ELISA applications, evaluate pairing with a suitable detection antibody and validate in the relevant sample matrix.
  • Cross-reactivity with mouse and rat is reported; confirm performance in the species of interest using appropriate positive controls.
