Biological Background: AIF Function and Localization

  • AIFM1 encodes apoptosis-inducing factor 1 (AIF), a mitochondrial flavoprotein with dual roles in metabolism and apoptosis. Multiple isoforms are produced by alternative splicing, including a brain-specific variant and isoform 5 that is frequently down-regulated in human cancers.
  • AIF functions as an NADH oxidoreductase, playing a critical role in mitochondrial respiratory chain biogenesis through interaction with CHCHD4 (PubMed:26004228). Related references: PMID:26004228
  • In response to apoptotic stimuli, AIF is released from the mitochondrion into the cytosol and nucleus, where its soluble form (AIFsol) induces caspase-independent DNA fragmentation (parthanatos) (PubMed:20362274). Related references: PMID:20362274
  • AIF also promotes apoptosis by binding EIF3G, inhibiting protein synthesis, and activating caspase-7 (PubMed:17094969). Related references: PMID:17094969
  • AIF plays a critical role in caspase-independent pyknotic cell death triggered by hydrogen peroxide exposure (PubMed:19418225). Related references: PMID:19418225
  • The nuclear form of AIF binds DNA in a sequence-independent manner, contributing to chromatin condensation during apoptosis (PubMed:27178839). Related references: PMID:27178839
  • AIF is widely expressed across tissues (PubMed:16644725), with protein detected in muscle, skin fibroblasts (PubMed:23217327), and osteoblasts (PubMed:28842795). Related references: PMID:16644725 PMID:23217327 PMID:28842795

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 450–550 of human AIF, a region highly conserved in mouse and rat, supporting the observed cross-reactivity.
  • For Western blotting, the predicted molecular weight of AIF is approximately 67 kDa; consider using appropriate positive controls such as HeLa or HEK293 cell lysates.
  • In immunohistochemistry (IHC-P), antigen retrieval optimization may be required; validate staining in tissues known to express AIF, such as muscle or skin.
  • For ELISA, use as a capture or detection antibody in sandwich ELISA development.
