Biological Background: AIF Function and Localization

  • The full-length AIF functions as an NADH oxidoreductase and, upon apoptotic stimulation, is released from mitochondria to the cytosol and nucleus, where its soluble form AIFsol binds DNA and induces parthanatos (caspase-independent chromatin fragmentation) (PMID:20362274, PMID:27178839).   • AIF interacts with EIF3G to inhibit protein synthesis and activates caspase-7, amplifying apoptosis (PMID:17094969). Related references: PMID:17094969

  • Consider titrating the antibody for optimal performance in immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P); perform appropriate antigen retrieval for IHC-P.
  • Validate antibody specificity using positive controls (e.g., HeLa cells) and knockout/knockdown models where possible.
