Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 619-848) of human AHR, which includes the transactivation domain. Consider its suitability for detecting full-length protein and potential isoforms.
- For Western blot, a band around 96 kDa is expected based on the target molecular weight. Validate specificity using positive and negative cell lysates.
- IHC-P and IF/ICC can detect both cytoplasmic and nuclear localization. Use known positive tissues (e.g., liver, lung) and include no-primary antibody controls.
- ELISA may be used for quantifying AHR in lysates; optimize coating conditions and pair with a validated detection antibody.
