Biological Background: AHR Function and Localization
- AHR (aryl hydrocarbon receptor) is a ligand-activated transcription factor of the basic helix-loop-helix (bHLH) family (class E bHLHe76) that senses environmental, dietary, microbiome, and metabolic compounds, playing critical roles in development, immunity, and cancer.
Related references: PMID:23275542 PMID:30373764 PMID:32818467 - Upon ligand activation, AHR translocates from the cytoplasm to the nucleus, heterodimerizes with ARNT, and binds to xenobiotic response elements (XRE) to drive transcription of target genes, including the xenobiotic-metabolizing enzyme CYP1A1.
Related references: PMID:23275542 PMID:30373764 PMID:7961644 PMID:33193710 - AHR mediates the biochemical and toxic effects of halogenated aromatic hydrocarbons and regulates angiogenesis, hematopoiesis, drug/lipid metabolism, cell motility, and immune modulation.
Related references: PMID:12213388 PMID:34521881 PMID:7961644 - Endogenous ligands, particularly tryptophan derivatives such as indoles and kynurenic acid, activate AHR to suppress adaptive anti-tumor immunity and promote cancer cell motility.
Related references: PMID:32818467 PMID:32866000 - AHR regulates the circadian clock by inhibiting CLOCK-BMAL1-mediated transcriptional activation of PER1, thereby repressing the basal and circadian expression of PER1.
Related references: PMID:28602820 - AHR is widely expressed across human tissues, including blood, brain, heart, kidney, liver, lung, pancreas, skeletal muscle, and retinal photoreceptors.
Related references: PMID:29726989 - Subcellular localization: AHR is primarily cytoplasmic under basal conditions but translocates to the nucleus upon ligand activation.
- Post-translational modifications: AHR is subject to acetylation and ADP-ribosylation, which modulate its transcriptional activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 619-848 at the C-terminal region of human AHR; the antibody may recognize both full-length AHR and C-terminal fragments.
- For Western blot analysis, a primary band near 96 kDa is expected. Validate reactivity in the relevant human sample system using known positive and negative controls.
- For ELISA applications, consider pairing this antibody with another monoclonal antibody targeting a distinct epitope to develop a sandwich ELISA. Optimize concentrations and dilutions experimentally.
- Cross-reactivity is reported for human AHR; if working with other species, verify cross-reactivity empirically due to potential sequence variability.
