Biological Background: AHCYL1 Function and Localization
- AHCYL1 encodes S-adenosylhomocysteine hydrolase-like protein 1, also known as IRBIT (IP3Rs binding protein released with IP3), DCAL, and putative adenosylhomocysteinase 2. Alternative splicing produces multiple isoforms.
- Acts as a multifaceted cellular regulator coordinating epithelial HCO3(-) and fluid secretion, mRNA processing, and DNA replication.
- Regulates intracellular Ca2+ signaling by interacting with ITPR1 to control ER-mitochondria Ca2+ transfer and apoptosis, in cooperation with BCL2L10. Related references: PMID:27995898
- Functions as an S-adenosyl-L-homocysteine (SAH) sensor that inhibits autophagy by binding to PIK3C3 and reducing phosphatidylinositol-3-phosphate production, independent of MTORC1. Related references: PMID:33993848
- Localizes to the endoplasmic reticulum, cytosol, apical cell membrane, and microsomes.
- Expressed in dendritic cells.
- Phosphorylation status modulates its inhibitory activity on ITPR1 and its interactions with CFTR, SLC26A6, and other binding partners.
- In vitro, lacks S-adenosyl-L-homocysteine hydrolase activity, distinguishing it from the related enzyme AHCY.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 31-260 of human AHCYL1; this region may recognize multiple isoforms. Validate antibody specificity in your target tissue or cell type.
- For Western blot, the predicted molecular weight is approximately 59 kDa. Optimize blocking and antibody dilution for your specific experimental conditions.
- Based on sequence homology, cross-reactivity with mouse and rat AHCYL1 is expected, but validation in each species is recommended.
- For ELISA, titrate the antibody against the antigen to determine optimal working concentrations.
