- It catalyzes the exchange of the acyl chain in acyl-dihydroxyacetonephosphate (acyl-DHAP) for a long chain fatty alcohol, yielding alkyl-dihydroxyacetonephosphate (alkyl-DHAP), the first ether-linked intermediate in ether lipid biosynthesis.
- The enzyme is located on the peroxisome membrane and within the peroxisome, reflecting its role in peroxisomal lipid metabolism.
- AGPS is a flavoprotein that uses FAD as a cofactor and belongs to the transferase class of enzymes.
- Mutations in AGPS are associated with rhizomelic chondrodysplasia punctata type 3, a peroxisomal disorder characterized by dwarfism, cataracts, and skeletal dysplasia.
- Post-translational modifications include acetylation and phosphorylation, as identified by proteomics studies.
