- AGMAT (guanidino acid hydrolase) is also known as mitochondrial arginase, guanidinobutyrase, and guanidinopropionase, reflecting its ability to hydrolyze various guanidino substrates.
- It catalyzes the hydrolysis of linear guanidino acids, such as taurocyamine, guanidinopropionic acid, and guanidinobutyric acid, into urea and corresponding amines, playing a critical role in guanidino acid detoxification.
- The enzyme shows substrate preference for molecules with a negatively charged head group and short side chains; it also metabolizes L-arginine with low efficiency.
- AGMAT is localized to the mitochondrion, directed by an N-terminal transit peptide, where it exerts its metabolic functions.
- Tissue expression is highest in liver and kidney, with moderate levels in skeletal muscle, brain, testis, skin, and the gastrointestinal tract. In the brain, regional differences exist with higher expression in the cerebral cortex than in medulla or spinal cord.
