Biological Background: AGER Function and Localization
- AGER (RAGE) is a cell surface pattern recognition receptor of the immunoglobulin superfamily that recognizes advanced glycation end products (AGEs), S100 proteins, HMGB1, amyloid-beta, nucleic acids, and histones. Related references: PMID:27572515 PMID:28515150 PMID:34743181
- A soluble isoform, sRAGE, generated by alternative splicing, acts as a decoy receptor that modulates ligand availability and signaling.
- Ligand binding triggers pro-inflammatory signaling via the adaptors TIRAP and MYD88, leading to NF-kB activation and production of cytokines IL6, IL8, and TNFα. Related references: PMID:21829704 PMID:33436632
- Mediates the cell membrane translocation of amyloid-beta peptide (ABPP) in cortical neurons, implicating it in Alzheimer’s disease pathogenesis. Related references: PMID:19906677
- Binds extracellular CpG DNA and promotes its endosomal uptake, activating intracellular inflammatory responses. Related references: PMID:24081950 PMID:28515150
- Facilitates non-professional phagocytosis of microbial spores and apoptotic cells, enhanced by binding to RNA, DNA, HMGB1, and histones. Related references: PMID:35974093
- Localizes to nuclear DNA double-strand breaks, enhances MRE11 endonuclease activity, and promotes DNA damage repair, supporting trophoblast invasion and placental maintenance. Related references: PMID:33918759
- Predominantly expressed in endothelial cells, with upregulated expression observed in pre-term labor and preeclampsia placentas.
Experimental Guidance and Technical Tips
- For Western blot, a band around 43 kDa is expected in human, mouse, or rat tissue lysates; reducing conditions and standard SDS-PAGE are recommended.
- In ELISA, the antibody can be used to detect AGER in serum, plasma, or cell culture supernatants; assay conditions should be optimized with a suitable antigen coating.
- The immunogen covers the extracellular domain (aa 30-330), so the antibody may recognize both membrane-bound and soluble forms; consider this when choosing sample types.
- Cross-reactivity with mouse and rat makes this antibody suitable for translational studies in these common model organisms.
