Biological Background: AGER (RAGE) Function and Localization
-
RAGE is a multiligand pattern recognition receptor that senses endogenous stress signals including advanced glycation end products (AGEs), HMGB1, S100 proteins, amyloid-beta, and histones. Related references: PMID:27572515 PMID:28515150 PMID:34743181
-
Ligand binding triggers pro-inflammatory signaling via NF-kappa-B, leading to induction of IL6, IL8, and TNFalpha. Related references: PMID:21829704 PMID:33436632
-
RAGE contributes to the pathogenesis of diabetes complications by mediating AGE-induced vascular inflammation. Related references: PMID:21565706
-
It participates in amyloid-beta peptide transcytosis across the blood-brain barrier in cortical neurons. Related references: PMID:19906677
-
Functions in DNA damage repair by localizing to double-strand breaks and enhancing MRE11 endonuclease activity; also protects replication forks from stress. Related references: PMID:33918759 PMID:36807739
-
Mediates endothelial albumin transcytosis via the RAGE/SRC/Caveolin-1 pathway, contributing to vascular hyperpermeability. Related references: PMID:27572515
-
Subcellularly localizes to cell membrane, early endosomes, and nucleus; a soluble form (sRAGE) is secreted.
-
Tissue specificity: predominantly expressed in endothelial cells; expression increased in pre-term labor placentas.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 40–210), which contains the ligand-binding region. This design is likely to generate antibodies that recognize cell surface RAGE.
- For Western blot, the predicted molecular weight is 43 kDa; however, glycosylation may cause migration at a higher apparent molecular weight. Consider validating in the relevant sample system.
- Suitable for ELISA, which may detect soluble RAGE (sRAGE) secreted into cell culture supernatants or serum.
- Cross-reactivity with mouse and rat enables translational studies. Verify cross-reactivity in these species by sequence alignment and pilot experiments.
