Biological Background: AGER Function and Localization
- AGER encodes the receptor for advanced glycation end products (RAGE), a pattern recognition receptor that senses a broad repertoire of endogenous stress signals, including AGEs, S100 proteins, HMGB1, amyloid-beta, nucleic acids, histones, and phospholipids. Related references: PMID:27572515 PMID:28515150 PMID:34743181
- Upon ligand binding, RAGE transduces pro-inflammatory signals via TIRAP and MYD88 adapters, leading to NF-kappa-B activation and induction of cytokines IL6, IL8, and TNFalpha. Related references: PMID:21829704 PMID:33436632
- RAGE mediates endothelial albumin transcytosis and HMGB1‑driven hyperpermeability through the RAGE/SRC/Caveolin-1 pathway, and loads HMGB1 into extracellular vesicles that promote hepatocyte pyroptosis via NLRP3 inflammasome activation. Related references: PMID:27572515 PMID:34743181
- The protein localizes to the cell membrane, early endosomes, and nucleus, and a secreted isoform exists; it is widely expressed on endothelial cells and is upregulated in pre‑term labor and preeclampsia placentas. Related references: PMID:33918759
- RAGE enhances DNA damage repair by interacting with the MRE11 complex at double-strand breaks, promoting end resection, and safeguards replication forks under stress by stabilizing the MCM2‑7 complex. Related references: PMID:33918759 PMID:36807739
- It enables non‑professional phagocytosis of spores and apoptotic cells by binding to RNA, DNA, HMGB1, and histones on the target surface. Related references: PMID:35974093
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 24‑344), which may capture soluble RAGE (sRAGE) as well as full‑length receptor in samples.
- For Western blotting, a band around 43 kDa is expected; consider using reducing conditions and validating in endothelial or inflammatory cell models.
- For IHC‑P/IF‑P, antigen retrieval may be necessary; test in positive control tissues such as lung or placenta where RAGE expression is documented.
- In ELISA, the antibody may be suitable for detecting sRAGE in serum or plasma; standardize coating and blocking steps according to the assay format.
