Biological Background: G Protein-Coupled Receptor Kinase 3 (ADRBK2/GRK3) Function and Localization
- ADRBK2/GRK3, also known as Beta-adrenergic receptor kinase 2 (Beta-ARK-2), is a serine/threonine-protein kinase that specifically phosphorylates the activated forms of G protein-coupled receptors (GPCRs).
- Phosphorylation of agonist-occupied beta-adrenergic and closely related receptors initiates beta-arrestin-mediated receptor internalization and desensitization, attenuating downstream signaling.
- Additionally, GRK3 phosphorylates ligand-bound anaphylatoxin receptors C3AR1 and C5AR1, leading to receptor desensitization. Related references: PMID:21799898
- GRK3 positively regulates the ciliary smoothened (SMO) signaling pathway by mediating phosphorylation of SMO, facilitating SMO trafficking into the cilium and stimulating SMO activity (By similarity).
- Subcellularly, GRK3 is localized at both postsynaptic and presynaptic compartments, highlighting its role in modulating neurotransmitter receptor signaling at synapses.
- Key functional keywords include ATP-binding, nucleotide-binding, kinase, transferase, and synapse, underscoring its enzymatic activity and synaptic involvement. The protein is also subject to ubiquitin-like conjugation (Ubl conjugation).
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1–300 of human ADRBK2, targeting the N-terminal region, which may recognize multiple isoforms or phosphorylation states.
- For Western blot (WB), expect a predominant band near 80 kDa in appropriate lysates; verify specificity using knockout or knockdown controls.
- In immunohistochemistry paraffin (IHC-P) applications, antigen retrieval and optimized antibody dilution are suggested; tissue-specific expression should be confirmed using known positive controls.
- The polyclonal nature of this antibody may offer enhanced sensitivity due to recognition of multiple epitopes; consider cross-adsorption if higher specificity is required for closely related GRK family members.
- For ELISA, the antibody can be explored as a capture or detection reagent; pairing with a matched detection antibody may improve assay performance.
