Biological Background: ADRB2 Function and Localization
- ADRB2 is a beta-2 adrenergic receptor, a G protein-coupled receptor (GPCR) that binds catecholamines epinephrine and norepinephrine, with ~30-fold higher affinity for epinephrine. Related references: PMID:2831218 PMID:33093660 PMID:7915137
- It couples to both G(s) and G(i) proteins, activating bifurcated signaling pathways: G(s) stimulates adenylyl cyclase/PKA, while G(i) activates PI3K/Akt promoting cardioprotective effects under hypoxia/oxidative stress. Related references: PMID:2831218 PMID:7915137
- In cardiomyocytes, ADRB2 regulates contraction rate; epinephrine induces rapid contraction via G(s)/PKA, while norepinephrine-mediated activation leads to slower contraction.

- In immunofluorescence/ICC, optimize permeabilization to access the intracellular epitope region (aa 300-400) as ADRB2 is a transmembrane protein.
- For ELISA, the antibody may be suitable for both direct and sandwich formats; validate with recombinant protein or peptide.
