Biological Background: ADRA1A Function and Localization

  • Alpha-1A adrenergic receptor is a G protein-coupled receptor binding catecholamines (norepinephrine, epinephrine), signaling primarily via G(q)/G(11) to activate phospholipase C and elevate intracellular calcium (PubMed:18802028, PubMed:37563160, PubMed:7815325). Related references: PMID:18802028, PMID:37563160, PMID:7815325
  • It can couple to G(14) and, in the nucleus, forms heterooligomers with ADRA1B to modulate phenylephrine-stimulated ERK signaling in cardiac myocytes (PubMed:18802028, PubMed:22120526). Related references: PMID:18802028, PMID:22120526
  • At the plasma membrane, interaction with CAVIN4/MURC regulates ERK activation in cardiomyocytes, contributing to cardiac hypertrophy (PubMed:24567387). Related references: PMID:24567387
  • Functions as a vasopressor in resistance arteries, playing a role in maintaining normal arterial blood pressure.
  • Localizes to the nucleus membrane, cell membrane, cytoplasm, and caveolae; post-translational modifications include disulfide bonds, glycosylation, palmitoylation, and phosphorylation.
  • Expressed in heart, brain, liver, and prostate; isoform 4 is the most abundant isoform in prostate, with high levels also in liver and heart.
  • Alternative splicing generates multiple isoforms; the receptor belongs to the G protein-coupled receptor family with seven transmembrane helices.

Experimental Guidance and Technical Tips

  • The immunogen sequence (aa 346-475, C-terminal region) may provide better specificity for ADRA1A, particularly for isoforms that share this region. Consider verifying reactivity with the relevant isoforms in the target sample system.
  • For Western blot (WB), a band around 51 kDa is expected based on the predicted molecular weight, though post-translational modifications may alter migration. Validate with appropriate positive controls (e.g., prostate or heart tissue lysates).
  • In immunohistochemistry (IHC) and immunofluorescence (IF/ICC), take into account the receptor's presence at the plasma membrane, nucleus, and cytoplasm. Optimize permeabilization conditions and validate with known expression profiles.
  • ELISA applications can leverage the antibody’s ability to capture native or recombinant ADRA1A. Cross-adsorption steps may be necessary to minimize background, especially when using complex samples.
  • The antibody's cross-reactivity with mouse and rat makes it suitable for translational studies in rodent models. However, always confirm cross-reactivity in the specific experimental context, as epitope conservation may vary between species.
