Biological Background: ADM Function and Localization

  • ADM, also known as AM or PAMP, is a multifunctional peptide hormone encoded by the ADM gene. It is produced as a precursor protein, pro-adrenomedullin, which is cleaved to generate several active peptides including adrenomedullin and PAMP.
  • ADM exerts potent hypotensive and vasodilatory effects, primarily through interactions with the CALCRL-RAMP2 and CALCRL-RAMP3 receptor complexes. Related references: PMID:8387282, PMID:9620797, PMID:32296767
  • In addition to vascular actions, ADM and PAMP regulate fluid and electrolyte balance, exhibiting diuretic and natriuretic properties in the kidney and inhibiting aldosterone secretion from the adrenal glands. Related references: PMID:8387282, PMID:9620797
  • Both peptides suppress basal ACTH secretion in the pituitary gland and act in the central nervous system to promote plasma volume loss, complementing their peripheral hypotensive actions. Related references: PMID:8387282, PMID:9620797
  • ADM also inhibits catecholamine release from sympathetic nerve endings and adrenal chromaffin cells, and serves as a ligand for MRGPRX2 receptors on mast cells, suggesting roles in neuroendocrine and immune modulation. Related references: PMID:10588445, PMID:9578982, PMID:15823563, PMID:34789875
  • ADM is primarily secreted and shows highest expression in pheochromocytoma, adrenal medulla, lung, ventricle, and kidney tissues. Subcellularly, it is secreted extracellularly.
  • Post-translational modifications include amidation, cleavage on paired basic residues, and formation of a disulfide bond, which are critical for its biological activity.

Experimental Guidance and Technical Tips

  • This antibody was raised against a synthetic peptide spanning amino acids 50-150 of human ADM. For Western blot, consider using denaturing and reducing conditions; the predicted molecular weight is approximately 20 kDa.
  • In ELISA, the antibody may be used as a capture or detection reagent, but optimal dilutions and conditions should be determined empirically in the relevant assay system.
  • Due to the secreted nature of ADM, appropriate sample types (e.g., serum, plasma, tissue lysates) and controls should be included. Cross-reactivity with mouse ADM has been observed, making it suitable for rodent models; however, validation in the specific experimental context is recommended.
