Biological Background: Adiponectin Receptor 1 Function and Localization
- Adiponectin receptor protein 1 (ADIPOR1, also designated PAQR1, TESBP1A) is a member of the progestin and adipoQ receptor (PAQR) family. It functions as a cell-surface receptor for the adipokine adiponectin.
- Receptor for ADIPOQ, an essential hormone secreted by adipocytes that regulates glucose and lipid metabolism; required for normal glucose and fat homeostasis and for maintaining a normal body weight. ADIPOQ binding activates a signaling cascade leading to increased AMPK activity, ultimately promoting fatty acid oxidation, glucose uptake, and decreased gluconeogenesis. Related references: PMID:12802337 PMID:25855295
- ADIPOR1 is widely expressed, with highest levels in heart and skeletal muscle (PubMed:12802337). Intermediate expression is found in brain, spleen, kidney, liver, placenta, lung, and peripheral blood leukocytes; weak expression in colon, thymus, and small intestine (PubMed:12802337, PubMed:16044242). Related references: PMID:12802337 PMID:16044242
- Subcellular localization is at the cell membrane, consistent with its role as a transmembrane receptor. The protein contains multiple transmembrane helices and a zinc-binding site, as indicated by structural studies (KEYWORDS: Transmembrane, Transmembrane helix, Zinc, Metal-binding).
- ADIPOR1 is involved in fatty acid and lipid metabolism, linking its function to metabolic diseases such as type 2 diabetes and obesity. Its signaling promotes insulin-sensitizing effects through AMPK and may protect against metabolic syndrome.
- Post-translational modifications and interaction partners remain under active investigation; however, the N-terminal region (used in this immunogen) is extracellular and critical for adiponectin binding.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular domain (amino acids 1-59) of human ADIPOR1, which is expected to recognize the native, membrane-bound receptor in denaturing and native assays.
- For Western blot, consider using reducing conditions and validate detection in lysates from tissues known to express ADIPOR1 (e.g., heart, skeletal muscle, or transfected cell lines). The predicted molecular weight is 43 kDa, but post-translational modifications or membrane extraction conditions may influence migration.
- For ELISA, the antibody can be used for capture or detection after appropriate optimization; pairing with another anti-ADIPOR1 antibody may improve sensitivity.
- Cross-reactivity with mouse and rat has been reported, making this antibody suitable for rodent models of metabolic disease. Species-specific sequence alignment (N-terminal region) confirms high homology, but validation in the relevant experimental system is recommended.
