Biological Background: ADHFE1 Function and Localization
- ADHFE1 (alcohol dehydrogenase iron-containing protein 1), also known as HOT, ADH8, or HMFT2263, is a mitochondrial hydroxyacid-oxoacid transhydrogenase.
- Catalyzes the cofactor-independent reversible oxidation of gamma-hydroxybutyrate (GHB) to succinic semialdehyde (SSA), coupled to reduction of 2-ketoglutarate (2-KG) to D-2-hydroxyglutarate (D-2-HG).
- Shows lower activity with substrates D,L-3-hydroxyisobutyrate and L-3-hydroxybutyrate (L-3-OHB), with approximately 10-fold reduced efficiency compared to GHB.
- Localized to the mitochondrion, consistent with its role in central metabolism.
- Expression is restricted to adult liver, indicating a tissue-specific metabolic function.
- Post-translational modifications include acetylation and phosphorylation, as identified by proteomics studies.
