Biological Background: ADH7 Function and Localization
- ADH7, also known as alcohol dehydrogenase class 4 mu/sigma chain, gastric alcohol dehydrogenase, or retinol dehydrogenase, is a zinc-binding enzyme belonging to the alcohol dehydrogenase family.
- Catalyzes the NAD-dependent oxidation of all-trans-retinol, alcohol, and omega-hydroxy fatty acids, with a preference for all-trans-retinol, 2-hexenol, and long-chain omega-hydroxy fatty acids such as juniperic acid; also capable of reducing all-trans-retinal and aldehydes in vitro. Through these activities, it plays a critical role in retinoid metabolism, fatty acid omega-oxidation, and the elimination of cytotoxic aldehydes produced by lipid peroxidation.

- The immunogen covers the full-length human ADH7 protein (aa 1-386), so the polyclonal antibody may recognize multiple epitopes and could detect both native and denatured forms.
- For Western blotting, a band corresponding to ~41 kDa is expected; however, validate molecular weight markers and consider possible post-translational modifications that may shift migration.
- For immunofluorescence, anticipate cytoplasmic staining; validate with appropriate controls and consider using a nuclear counterstain.
- Cross-reactivity has been reported for mouse and rat; optimize species-specific detection conditions for each experimental model.
- Given the preferential stomach expression, stomach tissue lysates or sections are recommended as positive controls; validate in the relevant sample system.
