Biological Background: ADH5/GSNOR Function and Localization
- ADH5, also known as alcohol dehydrogenase class-3, glutathione-dependent formaldehyde dehydrogenase (FDH, FALDH), or S-nitrosoglutathione reductase (GSNOR), belongs to the zinc-containing alcohol dehydrogenase family.
- Catalyzes the oxidation of long-chain primary alcohols and the oxidation of S-(hydroxymethyl) glutathione. Related references: PMID:8460164
- Also oxidizes long chain omega-hydroxy fatty acids, such as 20-HETE, producing both the intermediate aldehyde, 20-oxoarachidonate, and the end product, a dicarboxylic acid. Related references: PMID:16081420
- Class-III ADH is remarkably ineffective in oxidizing ethanol. Related references: PMID:8460164
- Required for clearance of cellular formaldehyde, a cytotoxic and carcinogenic metabolite that induces DNA damage. Related references: PMID:33355142
- Acts as a S-nitroso-glutathione reductase by catalyzing the NADH-dependent reduction of S-nitrosoglutathione, thereby regulating protein S-nitrosylation. (By similarity)

- Since the immunogen encompasses the full-length ADH5/GSNOR protein (amino acids 1-374), this antibody may recognize total ADH5 levels; consider using appropriate positive and negative controls.
- For Western blotting, a band around 40 kDa is expected; confirm specificity by using relevant cell or tissue lysates.
- In immunofluorescence applications, anticipate cytoplasmic staining in line with ADH5 subcellular localization; validate in your target sample system.
- For ELISA, optimize coating antigen concentration and antibody dilution to match assay requirements; cross-reactivity with mouse and rat ADH5 has been observed.
