Biological Background: ADH1B Function and Localization
- ADH1B, also known as alcohol dehydrogenase 1B or subunit beta, is an all-trans-retinol dehydrogenase belonging to the alcohol dehydrogenase family.
- Participates in retinoid metabolism by catalyzing the NAD-dependent oxidation of all-trans-retinol and its derivatives, and can also mediate the NADH-dependent reduction of all-trans-retinal derivatives. Related references: PMID:15369820, PMID:16787387
- Shows higher catalytic efficiency in the oxidative direction and displays similar affinity for all-trans-4-hydroxyretinol and all-trans-4-oxoretinal. Related references: PMID:15369820, PMID:16787387
- Subcellularly localized to the cytoplasm.
- A zinc-dependent and NAD-binding oxidoreductase.
- Subject to acetylation and phosphorylation, as detected in proteomic studies.
- Multiple isoforms can be generated through alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen comprises the C-terminal region (amino acids 206–375) of human ADH1B; epitopes within this fragment may be conserved across species, as evidenced by the reported cross-reactivity with rat ADH1B.
- For Western blot applications, a denaturing SDS-PAGE under reducing conditions is typically appropriate; the expected protein weight is approximately 40 kDa.
- The antibody has been validated for WB and ELISA; users should optimize dilutions and validate performance in their specific sample matrix and assay setup.
