Biological Background: ADCY2 Function and Localization
- ADCY2 encodes adenylate cyclase type 2, a member of the adenylyl cyclase family, which catalyzes the conversion of ATP to cyclic AMP (cAMP) upon activation by G protein-coupled receptors (PubMed:15385642). Related references: PMID:15385642
- The enzyme functions in signaling cascades downstream of muscarinic acetylcholine receptors and contributes to the regulation of gene expression, including increased IL6 production (By similarity).
- Alternative splicing generates multiple transcript variants, indicating potential isoform diversity (based on UniProt keyword).
- Subcellularly, ADCY2 localizes to the cell membrane and cytoplasm, consistent with a transmembrane protein that cycles between compartments.
- Post-translational modifications include phosphorylation (phosphoprotein) and N-linked glycosylation (glycoprotein), which may regulate activity and stability.
- Tissue expression is highest in the brain, particularly the caudate nucleus, cerebellum, and hippocampus; at the protein level, it is also detected in the zona glomerulosa and zona fasciculata of the adrenal gland (PubMed:11549699). Related references: PMID:11549699
Experimental Guidance and Technical Tips
- For Western blotting, a band around 124 kDa is expected. Use whole cell lysates from brain or adrenal tissues (human, mouse, rat) known to express ADCY2.
- For ELISA, validate the antibody’s specificity using recombinant ADCY2 protein or cell lysates from appropriate species.
- Consider optimizing antibody dilution and incubation conditions for each application, as performance can vary with sample type and assay format.
- Since ADCY2 is a transmembrane protein, use lysis buffers containing detergents (e.g., 1% NP-40 or Triton X-100) to ensure efficient solubilization.
