Biological Background: ADCY10 Function and Localization
- ADCY10 (adenylate cyclase type 10) is a soluble adenylyl cyclase (sAC) also designated SAC, HCA2, or testicular soluble adenylyl cyclase. Alternative splicing generates multiple protein isoforms.
- It acts as a bicarbonate and CO₂ sensor, catalyzing the formation of the second messenger cAMP from ATP (PubMed:12609998, 15659711, 24567411).
Related references: PMID:12609998, PMID:15659711, PMID:24567411 - ADCY10 plays an essential role in spermatogenesis: it generates cAMP that activates nuclear factors required for sperm maturation in the epididymis and triggers capacitation prior to fertilization.
- It is also involved in the regulation of ciliary beat frequency in airway epithelial cells (PubMed:17591988).
Related references: PMID:17591988 - Protein expression is highest in testis and airway epithelial cells; it is detectable at lower levels in multiple tissues including brain, heart, kidney, liver, lung, and pancreas.
- Subcellularly, ADCY10 localizes to the cell membrane, cytoplasm, cytoskeleton, perinuclear region, nucleus, cilium, and mitochondria.
- The enzyme requires divalent cations (Mg²⁺ or Mn²⁺) for activity and possesses ATP‑binding sites; several alternatively spliced variants exist.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N‑terminal region (amino acids 1–290) of human ADCY10; the antibody may recognize full‑length and some truncated isoforms.
- For Western blotting, the predicted molecular weight is 187 kDa. Use a 6–8% SDS‑PAGE gel and appropriate high‑molecular‑weight markers to achieve optimal separation. Consider extended transfer times for such a large protein.
- For ELISA, preliminary checkerboard titrations against recombinant or native ADCY10 are advised to determine optimal coating and antibody concentrations.
- The antibody has documented reactivity with human and mouse samples; validate performance in your experimental system, particularly if working with weakly expressing tissues.
