Biological Background: ADAMTS7 Function and Localization
- ADAMTS7 (A disintegrin and metalloproteinase with thrombospondin motifs 7) is a secreted metalloprotease that belongs to the ADAMTS family of extracellular proteases. It contains thrombospondin type-1 motifs and is synthesized as a zymogen requiring proteolytic activation.
- ADAMTS7 was initially reported to degrade cartilage oligomeric matrix protein (COMP), a key component of cartilage, suggesting a role in extracellular matrix remodeling and tissue morphogenesis. However, a more recent study found no proteolytic activity against COMP, indicating that its substrate specificity may be more restricted or context-dependent. - The protein is localized to the extracellular matrix and is secreted into the extracellular space, consistent with its function in modulating matrix composition and cell-matrix interactions.
- ADAMTS7 is broadly expressed in tissues, including heart, brain, placenta, lung, liver, skeletal muscle, kidney, pancreas, as well as in musculoskeletal tissues such as meniscus, bone, tendon, cartilage, synovium, fat, and ligaments.
- It is characterized by several structural features: a signal peptide, a prodomain, a metalloproteinase domain, a disintegrin-like domain, and multiple thrombospondin type-1 repeats, along with a C-terminal proteoglycan domain. It binds zinc and requires calcium for activity.
- Post-translational modifications include proteolytic cleavage at pairs of basic residues for activation, disulfide bond formation, and glycosylation. These modifications are critical for its secretion and enzymatic function.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (residues 1600-1686) of human ADAMTS7, which may detect both full-length and cleaved forms. For WB, a low percentage gel (e.g., 6-8%) and extended transfer time are recommended due to the high molecular weight (~184 kDa).
- For ELISA, the antibody may be used as a capture or detection reagent; titrate to determine optimal working concentration.
- Given the polyclonal nature, validate performance in your specific sample matrix and include appropriate controls.
- Cross-reactivity is reported for human; if using samples from other species, verify reactivity experimentally.
