Biological Background: ADAMTS4 Function and Localization

  • ADAMTS4 (a disintegrin and metalloproteinase with thrombospondin motifs 4), also known as aggrecanase-1, is a zinc-dependent metalloprotease that cleaves aggrecan at the Glu373-Ala374 bond (also designated Glu392-Ala393).   • Additionally, ADAMTS4 cleaves cartilage oligomeric matrix protein (COMP), further contributing to extracellular matrix remodeling. Related references: PMID:39672391
  • The enzyme is secreted into the extracellular matrix where it mediates proteolysis.

Researchers should consider that this antibody may recognize both the proform and the mature enzyme after prodomain removal, depending on the experimental conditions.
  • For Western blot (WB), a band at approximately 90 kDa (full-length zymogen) is expected, with possible detection of processed forms at lower molecular weights.
  • This antibody is suitable for WB, IF/ICC, and ELISA. Cross-reactivity with mouse and rat ADAMTS4 has been observed, making it useful for studies in rodent models. However, we recommend validating the antibody in the specific sample system and assay format.
