- ADAM28 (Disintegrin and metalloproteinase domain-containing protein 28) is a member of the ADAM family, also known as eMDC II (epididymal metalloproteinase-like, disintegrin-like, and cysteine-rich protein II) or MDC-L. It is encoded by the ADAM28 gene (aliases ADAM23, MDCL) and undergoes alternative splicing.
- It functions as a metalloprotease involved in adhesive and proteolytic events during lymphocyte emigration. It may mediate ectodomain shedding of lymphocyte surface proteins such as FASL and CD40L, thereby modulating immune responses.
- ADAM28 may also play a role in sperm maturation, based on its expression and functional context.
- Subcellular localization includes the cell membrane and secretion, consistent with its transmembrane and secreted domains. The protein contains a signal peptide, propeptide, metalloprotease, disintegrin, cysteine-rich, EGF-like, and transmembrane domains.
- Key post-translational modifications include glycosylation, disulfide bond formation, and cleavage on a pair of basic residues to convert the zymogen into an active protease. It binds zinc as a metalloprotease cofactor.
- ADAM28 is predominantly expressed in secondary lymphoid tissues—lymph node, spleen, small intestine, stomach, colon, appendix, and trachea—with the lymphocyte population being the primary source. Isoform 2 shows preferential expression in the spleen.

- The immunogen spans amino acids 199-440 of human ADAM28, which lies within the extracellular region containing protease, disintegrin, and cysteine-rich domains. This design may recognize both the membrane-bound and secreted forms of the protein.
- For Western blot, consider using lysates from human lymphoid tissues (e.g., lymph node, spleen) or cultured lymphocytes, as these are known expression sites. Mouse cross-reactivity should be validated with appropriate tissue or cell line controls.
- SDS-PAGE sample preparation may require reducing conditions to resolve disulfide-linked structures; the predicted full-length molecular weight is ~87 kDa, though fragments may be observed due to zymogen processing.
- In ELISA applications, evaluate the detection of soluble ADAM28 in biological fluids or cell supernatants, as the protein can be secreted. Include recombinant protein or known positive controls for assay optimization.
