Biological Background: ACTR3B Function and Localization
- ACTR3B (Actin-related protein 3B), also known as ARP3-beta or ARP4, is a member of the actin-related protein family and shares homology with actin.
- It is proposed to function as an ATP-binding component of the Arp2/3 complex, playing a role in the regulation of actin polymerization and mediating the formation of branched actin networks.
- The protein contributes to the organization of the actin cytoskeleton and may influence cellular processes such as migration and metastasis.
- Subcellularly, ACTR3B localizes to the cytoplasm, cytoskeleton, and cell projections, consistent with its role in actin dynamics.
- Tissue specificity: Detected in fetal brain, adult brain (neurons in gray matter, but not white matter), liver, skeletal muscle, and pancreas. Notably, it is detected in lung adenocarcinoma cells with low metastatic potential, but not in those with high metastatic potential, suggesting a potential role in decreasing metastatic potential.
- Alternative splicing of ACTR3B gives rise to multiple isoforms, and the protein contains actin-binding and ATP-binding domains.
- The calculated molecular weight of the canonical isoform is approximately 47.6kDa, with the observed band around 48kDa.
Experimental Guidance and Technical Tips
- For Western blot analysis, a specific band around 48kDa is expected. Validate the antibody in your target tissue or cell line, considering that expression levels may vary (e.g., differential expression in metastatic versus non-metastatic cells).
- When using ELISA, optimize coating antigen concentration and detection conditions; ensure the recombinant protein immunogen matches the assay format.
- Cross-reactivity with mouse is reported, making this antibody suitable for rodent model studies. Confirm reactivity in your specific mouse strain if needed.
- Due to the polyclonal nature, lot-to-lot consistency should be verified in critical quantitative assays.
