Biological Background: ACTL6B Function and Localization
- ACTL6B (Actin-like protein 6B), also known as BAF53B, ArpNalpha, is a 53 kDa BRG1-associated factor encoded by the ACTL6B gene.
- It is a core component of the SWI/SNF chromatin remodeling complex, which alters DNA-histone contacts in an ATP-dependent manner to regulate transcription.
- ACTL6B is specifically incorporated into the neuron-specific nBAF complex, replacing ACTL6A during neural differentiation, and is essential for postmitotic neural development and dendritic outgrowth.
- The nBAF complex, together with CREST, targets promoters of genes critical for dendritic growth; studies show ACTL6B is indispensable for this process (PubMed:31031012). Related references: PMID:31031012
- Localized predominantly in the nucleus, consistent with its chromatin regulatory function.
- Mutations in ACTL6B are associated with developmental epileptic encephalopathy and intellectual disability, highlighting its role in neurodevelopment.
- Tissue expression is neuronal; it is required for dendritic maturation in postmitotic neurons.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1-280 amino acids of human ACTL6B; the resulting polyclonal antibody may recognize multiple epitopes within this region.
- For Western blotting, a band around 47 kDa (the predicted molecular weight) is expected in reduced samples; use appropriate molecular weight markers.
- In IHC-P and IF/ICC, expect nuclear staining; antigen retrieval may be required for formalin-fixed tissues.
- The antibody cross-reacts with mouse and rat ACTL6B, extending its utility to rodent models.
- For ELISA, consider pairing with a matched detection antibody if quantitative detection is needed, and validate in the relevant sample matrix.
