Biological Background: AP-2-alpha (TFAP2A) Function and Localization

• AP-2-alpha is a sequence-specific DNA-binding transcription factor that recognizes the consensus motif 5'-GCCNNNGGC-3', regulating viral and cellular enhancer elements.
• It activates genes essential for morphogenesis of the eye, face, body wall, limbs, and neural tube, and is the only AP-2 family member required for early lens vesicle development.
• AP-2-alpha also acts as a transcriptional repressor for genes such as MCAM/MUC18, C/EBP alpha, and MYC, and negatively regulates Wnt canonical signaling. Related references: PMID:33000225
• Together with CITED2 coactivator, it stimulates transcription from the PITX2 P1 promoter by associating with chromatin at the promoter region.
